Studies on the Pesticide Amitraz-induced Hypotension

Abstract
Amitraz is a formamidine pesticide widely used in veterinary medicine. It can produce various side effects that resemble those of the ¥á2-adrenoceptor agonists, such as xylazine and clonidine. In an attempt to explain some of its side effects, the hypotensive effect of amitraz was studied in urethane anesthetized rabbits. The intravenous injection of amitraz (0.5~2.0 mg/kg) to anesthetized rabbits caused a dose-dependent fall in blood pressure. The hypotensive effect of amitraz (1 mg/kg) was significantly prevented by the pretreatment of both yohimbine (2.0 mg/kg, i.v.) and atropine (2.0 mg/kg, i.v.), but not by the pretreatment of dl-propranolol (1 mg/kg, i.v.). The results obtained in the present study suggest that ¥á2-adrenoceptor mediates amitraz-induced hypotensive effects and amitraz also cause the hypotensive effect through an increased cholinergic activity.
